She graduated from Fort Hunt High School in 1982. She was chosen to speak at her graduating class's baccalaureate service and was crowned homecoming queen.

Kotb belonged to the Beta Nu Chapter of the Delta Delta Delta women's sorority during her time in college at Virginia Tech. Kotb received a Bachelor of Arts in broadcast journalism in 1986.

At her alma mater's 2008 Virginia Tech graduation, Kotb gave the keynote address and used her iPod to play Metallica's "Enter Sandman" during her speech.  Kotb was chosen in 2010 to serve a three-year term on the board of directors of the Virginia Tech Alumni Association.

Kotb wed Burzis Kanga, a former tennis instructor at the University of New Orleans, in 2005. In 2008, the couple got divorced.
In 2013, Kotb started dating New York financier Joel Schiffman.

Kotb made the announcement that she was engaged to Schiffman live on Today on November 25, 2019.

Kotb and Schiffman called off their engagement on January 31, 2022, and Kotb stated during the fourth hour of Today (Hoda & Jenna) that they would concentrate on co-parenting as friends going forward.

Later, Kotb worked as a reporter for the ABC affiliate WQAD in Moline, Illinois, from 1988 to 1989. From 1989 until 1991, she worked as a reporter and weekend anchor for the Fort Myers, Florida, CBS affiliate WINK.

Kotb worked as an anchor and reporter for the CBS affiliate WWL in New Orleans, Louisiana, from 1992 to 1998.

Kotb was requested to appear in Martina McBride's 2011 music video for the ballad "I'm Gonna Love You Through It," which is dedicated to cancer survivors.
